Practical Method
Mix amylase, starch, and a buffer solution of a specific pH.
Every 30 seconds, add a drop to iodine on a spotting tile.
Stop when the iodine remains orange (no starch left).
The independent variable is the pH; the dependent variable is the time taken for starch to disappear.
